My nissan elgrand e52 have transmission problem driving fine no noise at all or sign parked fine but when I started my journey after couple of hours and then car not moving but going in all modes.

You need to check auto transmission fluid level and top up if needed or change

I have checked auto transmission fluid level which is fine doesn't need changing but still having same problem but would also like to ask when is best to check auto transmission fluid level when engine is cold or warm.

Your transmission fluid should be red in colour and not discoloured or smelling burnt . If o.k you really need to get it scanned for fault codes
